Human airway air-liquid interface (ALI) cultures have been commonly used to model various mechanisms of coronavirus pathogenesis where the cells are efficiently infected by human or animal-transmitted CoV, including SARS-CoV, SARS-CoV-2, and MERS-CoV. Liquid handling is an important part of automation for HTS especially as the demand for standardization and efficiency increase. With cell based assays, liquid handling instrumentation can be used to seed, wash and assay multiple plates faster than manual methods and with less variance. Caliper’s Sciclone® ALH 3000 liquid handling instrument and LiCONiC® STX40 CO2 incubator were used to demonstrate that complex, multiple step cell based assays could be optimized for compatibility in a fully automated system using Corning® HTS Transwell® 96 permeable support plates. Drug transport and chemotaxis assays were seeded, incubated, washed and assayed using the Sciclone ALH 3000 instrument and LiCONiC STX40 incubator. Drug transport assays were evaluated through Transepithelial Electrical Resistance (TEER) measurements, Lucifer Yellow (LY) rejection and Rhodamine 123 efflux using the MDCK/ MDR1 cell line. Evaluation of manual versus automated handling showed similar cell monolayer integrity based on TEER and LY results and similar cell functioning based on Rhodamine 123 efflux. Evaluation of Chemotaxis assays were based on percent invasion of HT-1080 cells using serum as a chemoattractant as well as the ability for equal cell numbers to be seeded. Both manual and Sciclone data shows similar chemotaxis response based on conditions tested. Our results demonstrate that, with assay optimization, non homogenous cell based assays can be fully automated to yield highly reproducible data.

This poster compares the performance of the Corning Transwell 96 well based permeable support assay system to a 24 well format based process. It shows that chemotaxis, invasion and drug transport assays, performed in this high throughput configuration are equivalent to those done in the lower throughput 24 well systems. This allows a higher number of samples tested per day while using lower amounts of reagents per sample.
